We are looking for a talented Molecular Biologist to join our team and contribute to groundbreaking mRNA vaccine development. The ideal candidate will have experience in molecular and cell biology techniques, including DNA and RNA extraction, PCR, cloning methods, cell line maintenance, transfection, protein expression, and protein purification. You will play a critical role in advancing our mRNA and protein-based therapeutics pipeline, working across multiple areas of our research and development efforts.
Key Responsibilities:
- Perform DNA and RNA extraction and plasmid extraction at various scales.
- Execute vector transformation in bacterial strains such as DH5-Alpha or similar.
- Apply a wide range of PCR techniques including inverse, touch down/up, gradient, long amplicons, TaqMan, and real-time PCR for various applications.
- Carry out cloning via methods like cut-and-paste, Gibson assembly, Gateway, and In-Fusion.
- Handle, maintain, and stock various cell lines, particularly HEK 293T, HeLa, MCF-7, and suspension cell lines such as Expi and FreeStyle series.
- Perform transfection using both chemical and electroporation methods at varying scales.
- Achieve protein expression in bacteria, yeast, insect, and mammalian cells, and support protein separation and purification (optional).
- Conduct protein estimation assays (e.g., Bradford, BCA, or ELISA).
- Use Western blotting for estimating relative protein expression levels.
- Utilize FPLC systems such as Cytiva's Akta series or Bio-Rad's NGC line (desirable but not mandatory).
- Collaborate with cross-functional teams to advance various R&D; projects in mRNA vaccine development, cell line generation, and protein production.
Qualifications:
- PhD in Molecular Biology, Cell Biology, or a related field; OR a Master's degree in a relevant field with at least 3+ years of additional experience in a relevant laboratory environment.
- Hands-on experience with DNA and RNA extraction, as well as plasmid extraction at various scales.
- Extensive expertise in PCR techniques, including real-time PCR and TaqMan assays, for various molecular biology applications.
- Proficiency in cloning methods, including cut-and-paste, Gibson assembly, Gateway, and In-Fusion.
- Experience with cell line maintenance (especially HEK 293T, HeLa, and MCF-7) and suspension cell lines like Expi and FreeStyle.
- Expertise in transfection techniques, including chemical and electroporation methods.
- Demonstrated ability to achieve protein expression in bacteria, yeast, insect, and mammalian cells.
- Experience in protein separation and purification methods (e.g., FPLC).
- Familiarity with protein estimation assays like Bradford, BCA, or ELISA, and knowledge of Western blotting for protein expression analysis.
- Experience with FPLC systems (Cytiva's Akta or Bio-Rad's NGC series) is a plus but not mandatory.
- Strong ability to work independently and within a team environment, effectively managing experiments and troubleshooting when necessary.
Preferred Experience:
- Experience with mRNA IVT (in vitro transcription) techniques.
- Prior exposure to monoclonal antibody (mAb) production.
- Familiarity with cell banking and secure cell line generation.
- Experience working in a cGMP or regulated environment is a plus.
- A track record of publishing scientific findings and presenting them clearly in both written and oral formats.
