Media Preparation Sterilization

Prepare tissue culture media according to SOPs and crop-specific protocols.

Accurately weigh and dispense macro nutrients, micro nutrients, vitamins, sugars, agar, gelling agents, and plant growth regulators.

Monitor pH adjustment, media consistency, and gel strength.

Operate autoclaves, media dispensers, water purification systems, and laboratory equipment.

Ensure complete dissolution of chemicals and proper heating procedures before sterilization.

Maintain sterility throughout the media preparation process.

Media Optimization Troubleshooting

Understand the effect of nutrients, hormones, and gelling agents on culture performance.

Assist in optimizing media for multiplication, rooting, elongation, and hardening stages.

Identify causes of poor culture growth, contamination, vitrification, browning, weak rooting, or low multiplication rates.

Recommend adjustments in hormone concentrations, media composition, pH, agar concentration,



or sterilization procedures.

Support protocol development for banana, bamboo, blueberry, ornamentals, and other crops.

Quality Control

Monitor media batches for consistency and quality.

Maintain batch records and traceability of raw materials.

Ensure adherence to laboratory SOPs and quality standards.

Conduct periodic checks on stock solutions and chemical inventory.

Documentation

Maintain media preparation logs and production records.

Record observations and corrective actions.

Support laboratory audits and compliance requirements.
